W. Reiner 1952 Journal 23Feb. Calif.: Santa Clara Co., Stanford University Spent 1500-1630 hrs with Dr. V.C.Twitty with object of learning technique used in hybridizing Triturus species. Procedure: Use animals only where when actively amplying and spawning in the field. Perhaps 767, but not 99, store well at low temperatures of 3-4°C. Cut off head, slit belly from neck to vent, remove most of intestine. Snip out ovaries of one male and macerate in about 5 cc of Holtfreter's solution - solution need not be sterile. Snip out oviducts, placing them on glass surface moist with Holtfreter's. Free eggs by stripping or dissecting away oviduct. Place in sperm suspension for short time and remove to damp - not wet-glass bowl and cover. Add water after about 30 minutes. Pond water best. Not distilled or chlorinated tap water. Cleavage should occur within 12-18 hrs. Larvae photographed by following method: Glass container (sawed of salt shaker) with beeswax layer in bottom. Way grooved so that larvae when laid in it remains in fixed position. Container over filled with water and glass plate slid across surface excluding all air bubbles. Photographed then with camera vertical. Larvae previously anesthetized.
